The departure time is approximate, you can get on at any available time. The bus runs every 30 minutes starting at 10:20 a.m. for the line in Milan, and every 20 minutes starting at 9:00 a.m. in Rome. The departure point for the line in Rome is at Termini Station. From the time of obliteration, the ticket is valid for one year. You can use the service in Milan on a given day and in Rome on another of your choice, as long as it is within one year from the time of obliteration. You can also reverse the two cities, using the service first in Rome, then in Milan, with the same conditions mentioned above.
